Fuzzy has to go to university at eight o'clock so we wake up super early and prepare for a day of battle. Takes her to the bus stop and she is still mumbling that I am driving too slow! >_<
Go to Lye before anyone else and borrow key from the farm manager to get into the lab. Start experiment around 8:45 a.m. and shall finish soon. Still one more time point to go.
So many things to do! Actually is the busiest Friday ever. Will practice cello with UYO and then go on to pub crawl for WESA (engineering club). @_@ Hope will be ok tomorrow morning and tomorrow evening has to go out for dinner.

Lost souls
Watch Lost so I think I will do my diary and the TV description in two seprate blog sessions.
I suddenly understand that this TV program is talking about a bunch of lost souls. Tom Sawyer is lost not only because the airplane crashed, but he had killed the wrong man and hasn't really avenged his mother. Charlie has lost his dignity because he had no money and Ethan had taken away his freedom. He killed because he was insecure. Kate killed because she had to, but she regreted it immensely because she killed the one she loved the most. Jack has lost his father without saying goodbye. The kid lost his mother, and his father lost the years with him when he was young. Lock has gained the ability to walk, but noone loves him so he is a lost soul. Sayed lost his lover.
This episode deals with murders. I thought it wasn't LOST because we see a little boy and his mother is being murdered by a man. He hides under the bed so he can't find him.
Sawyer grows up. He finds the killer in Australia, and buys shrimp from him. However, before his death, Sawyer finds out that he is not the killer so essentially he has killed the wrong guy.
Charlie is dealing with post traumatic syndrome of killing Ethan. He works extremely hard on everything. Hurley buries Ethan with Charlie and he suggests to Sayed that he should talk with Charlie. He relates a story of killing a bomb maker in a firing squard and having nightmares after the kill. Charlie should talk about the incident with other people and not bottle up his feelings.
Sawyer meets Jack's father in a pub. He inadvertently urged Sawyer to kill his mother's killer by saying that he should not lie down and take life as fate. He finds out from Jack that his father said "That's why the Red Socks never wins, because it is fate." Jack doesn't believe in fate, but his father does and do nothing to it.
Kate hunts the boar with Sawyer, they meet Lock, who said that Sawyer shouldn't blame himself for his mother's death, and shouldn't blame the boar for dragging his tent and eating his food, and shouldn't blame himself for killing the wrong man.
It is an amazing program. These people all had a past that haunts them, in dreams and memories they remeber the good days, the bad days, the sad days, and the happy days of their former life. Even though they are living on the island together, they can't get away from the past. The past makes who they are now and all their actions are dictated by their past experience. That's what made LOST such an interesting program and that's why I love it.

Calling for pictures
Our biological secretary has called for pictures that has been taken in the fields and in labs. I don't have a digital camera, but work may have one so I will be able to take some pictures and save it and print it out in city. Cool.
Yesterday spend whole day running several experiments. So tired! Hand hurts after volleyball game but also because uses too much for pipetting. Write with left hand now because right hand is overused.
Mum cooked a delicious beef soup. It has potato, carrot, onion and beef. We drank many bowls of soup and watch the news with dinner. The foot-and-mouth thing is still treated as a hoax and no animal has been found to be sick at all. It probably is a hoax. However, we have to be careful never to bring such disease into the country.

The experiment
Hmm...I have never thought about how to say this but I will just come out and say it.
In the beginning I didn't really know what I was doing, but I have set out a plan for what I have to do so I just go out and do it.
My thesis consists of four main parts of experiment. The first part is the Oxidation-reduction potential experiment. Basically this is just using a oxidation-reduction potential (ORP) meter to measure the ORP, which is measured in mV, of a liquid. This is an indirect measure of the ozone concentration of the liquid. The second part of the experiment is using Indigo reagent to measure the concentration of ozone in liquid. This is to dilute the Indigo solid into liquid and ozone will bleach the substance, so the original color is blue and the high ozone concentration will make the solution white or transparent. The third part of the experiment is to inhibit the bacteria in laboratory, using blood esculin plates. This experiment will include gas exposure, i.e. put bacteria on plates then expose to ozone gas/ozone liquid; liquid exposure, i.e. expose bacteria in liquid to ozone straight, or add ozonated liquid to liquid bacteria then grow bacteria on blood esculin plates; solid: grow bacteria on blood plate and when it is dry, apply ozone or apply ozone to liquid and put filter paper in the liquid, put this piece of paper on the dry blood plate that has bacteria. The last part of the experiment will involve cow work, but this part is unlikely to be done now.
To describe my work further, have to tell you more. Ozone gas is not good news to people's lungs so better put it into solution. However, ozonated solution has a lower bactericidal ability to ozone gas. The Indigo and ORP experiments aim to measure the concentration of ozone in liquid (water mainly). The bacteria experiment aims to find the best way to use ozone to kill bacteria. Mastitis is caused mainly by bacteria and that is why I am using ozone to kill the bacteria.

Too many social events
I have been called late last evening by my friend saying that we should go listen to a cello concert next Thursday evening. I also have rock climbing that night. Wondering whether to go to both or not?
After work, I take my friend to the movie theatre to buy movie money for next week's Star Wars premier on midnight Wednesday. Wish everything will be ok. Too many social events! Maybe I should sleep before going to Star Wars and sleep again after the movie so I can get up and work on the next day. Too busy!
Work is going well but should use a pipette instead of a graduated cylinder to measure our 5 ml Triton X-100. It is a detergent, for God's sake, it sticks to every container that you put it into so you shouldn't put it in a container!
No plates in the farms so I won't be able to do any bacteria experiment until Friday.
Scare about Foot and Mouth disease out break in Waihiki Island, however until now it is still treated as a hoax. Vet will test the animals on the island and the stock will not be moved in or out of the island now. However, pets are not restricted.
If Food and Mouth is really here, then New Zealand will lose millions of dollars.
Australian rubgy league players are leaving Australia to play in Europe. Very sad. One coach loses players to injuries, tackle charges and oversea clubs. Sounds like he is not going to have enough players for the next test. Poor guy.
The Peter Dunn fiasco will not leave Helen Clark alone, but now she is fighting back saying that National is paying Peter's family to drag the dirty laundry out. I don't care. The whole thing is stupid and not about the ability she runs this country. Just like Winston Peters saying that ailing 70 years old Iraqi officials living in New Zealand is going to bomb us: that's outrageous and impossible. He can't even walk straight! He looks older than the Pope! How can he be a security risk? Winston looks like a laughing stock now. No body seems to notice how old the guy is and just focus he used to work the bad guys, but essentially, you need to be able to walk and move if you want to be a security risk.
Enough rambling. Should go back to the detergent experiment and keep my head down till boss comes back to find a very busy e-mail box. Hope this disease thing is not real otherwise we will face a bad economy.

Star Wars Episode I: The Phantom Menace
Watch Star Wars Episode I last night. Just wondering what they will do next year or the year after when even Episode III can be played on television? Doesn't that mean that we can go through from the Episode I to Episode VI? It will be the same experience by watching Lord of the Ring I to III, I suppose, but that means it will take the whole day, or three weeks for television to play it, and you have to wait a long time in between. Maybe it will be better to get DVD?
So many movies are out this week. Guess Who, The Interpreter, In Good Company, Kingdom of Heaven, and Bewitched is coming out soon. ^_^ I am glad that I bought the Movie Money thing because it is much cheaper than the normal ticket price. Too bad Fuzzy won't be able to watch Star War with us on the first night. I will tell her all about it when I get home.
Work is hectic. I have run two experiments today, one for RO @ 4 degree C, the other Citrate buffer 1% v/v. They both worked well but I have serious doubts for the ORP readings. They may still be high because the meter hasn't gone back to baseline. Will see what is happening today when I run another citrate buffer.
Before boss comes back I should carry out another bacteria experiment, both growth curve and the ozone experiments can be run tomorrow and on Thursday. I will be extra busy this week!
Research on the nonionic detergents that supervisor told me about. A lot of information to digest.
Don't know whether will go rock climbing today or not, but will definitely go to the meeting with ROC club tomorrow. Should post the wretch photoalbum site so people can see some rock climbing pictures. Cool. I will show the picture of me climbing up the wall. ^_^ Love rock climbing.

Internet connection
Mum and dad are chopping up our friend's tree, and I can finally get online without getting cut off. So happy! So I train Colin for a while and manage not to die! YA! This time I am no longer killing gnomes, I am obliterating boggart, banshee, and cap in the Green House in Hogwarts ground. Very happy. Also change into another character so I can follow Aeka to a group. I was a Bishop, doing all the healing for the team mates. So happy! I am not dumb anymore! Haha. Get 1 sword from the group for the person who owns the bishop to thank him for his generosity.
Tofushark comes to our house to repair her pants. Fuzzy gets the sewing machine out and they go on to repair two pants so she can wear them to university. We are eatting chestnuts. They bring back the cake tins and plates.
Mum and dad are back before dinner. I start making dinner when they go home. Chopping up beef, spring onion, carrots, bamboo shoots, and wood ears. Hands tired after that. Make microwave fried chicken, takes 10 minutes in total to cook. Sour hot soup is a success, and the stir-fry beef spring onion is also delicious. We have Fuzzy's jelly and fruit after dinner, and mum's red bean soup for night cap.
Watch CSI and Gill Grissm is demoted after Eckley's investigation. Katherine is supervising the team and Gill is working with Sarah and Sophia. However, this will not change CSI too much, just that Gill is no longer the supervisor of everyone. The Graveyard shift will never be the same without Grissm as leader, but I am sure the story will go on portraying what are happening in the crime lab to all the former members of the team of CSI.
Criminal Intent is about a woman who died after obtaining the little black book of a swinger who owns a porn calendar/magazine franchise. A candidate and his wife are on the little black book. They were in a sex club ten years ago. They have since divorced, but to protect their past and scandal, one of them killed the woman, and later the person who stole the little black book and blackmail them. We discover that the woman killed both persons in the end because she wants her daughter to think that she is not a bad mother.
Today I will do RO water at 4 degree C. Hope it will go well. Has to make up more Indigo stock solution first. Research for Triton, Tween, and MP.

Happy Mother's Day
I have tried to do some work over the weekend, but got completely bored after five files. Watch Thomas Crown Affair again this evening, but tomorrow will definitely watch a little bit of the dance with star on Sunday. Has bought the tofu from New Safe. Meet M's mum at New Safe and said hi. Mum and dad are helping a friend to chop up their branches and will move them to the recycling centre.
Tonight I am going to the chef. Happy Mother's Day.
